BRIEF CRITICAL VIEWS ON MY CONTRIBUTION TOWARDS THE ADVANCEMENT OF SCIENCES:

I did Ph.D. in Physiology from the University of Calcutta and his thesis (1993)was entitled “Metallic Pollution: Its Effect on a male reproductive system with special references to protein inadequacy”. My earlier research works basically on heavy metal toxicities and how these metals especially divalent cation alters system biology. My research established a link between nutritional role especially dietary protein and severity of impaired functional biology including reproductive health in experimental animals. I exclusively worked on changes in DNA, RNA, and genetic information systems due to heavy metal assaults in albino rats in relation to dietary proteins and antioxidant vitamins, particularly Vitamin C and E (Das & Dasgupta 1997; Das & Dasgupta 2000; Das & Dasgupta 2002; Das et al 2020). My team’s works on vitamin C and E against nickel, Cr VI and Pb treated pathophysiology of cardiovascular, renal, pulmonary, reproductive, and hepatic-biliary systems are noteworthy (Gupta 2008; Das 2015). Our works also showed an elevation of serum glucose level in rats supplemented with a megadose of vitamin C as l-ascorbic acid. Further of his research extended to human occupational exposure to heavy metals especially on lead (Pb) and oxidative stress though NF-kβ (Das et al 2011; Chadchan et al 2014; Mantur et al 2014; Das et al 2016; Das et al 2017).

Further our research on experimental biology progress on low oxygen microenvironment in rodents especially in chronic sustained and intermittent hypoxia in experimental animals treated with heavy metals (Nickel and Lead). These work outcomes showed cell signal transduction in hypoxia exposed animals in system biology though hypoxia sensitive genes like hypoxia-inducible factor-1α (HIF-1α), erythropoietin (EPO) and vascular endothelial growth factor (VEGF) expressions in presence of divalent cation metals (Hattiwale et al 2013; Das et al 2015; Reddy et al 2020). Further, we had done extensive research on the regulation of vascular endothelial and smooth muscle integrity though NOS3/NOS2 gene expression in hypoxia exposed rodents (Patil et al 2015; Das and Saha 2014). One of our works on hypoxia, divalent heavy metals, and glucose intolerance in rodents showed the definite involvement of L/N type of calcium channel and cardiac autonomic dysfunctions with both chronic sustained and intermittent hypoxia. This work suggests possible therapeutic uses of L/N type of calcium channel blockers to control hypoxia or metal-induced hyperglycemia. We have also engaged in research on aging-related oxygen sensing gene and vascular physiology relationship, especially on arterial stiffness including augmented index in humans (Das et al 2016, Das et al 2017; Khodnapur & Das 2021).

Our group invented a new modified method of estimation of serum Vitamin E (α-tocopherol) which is economic, less tedious, and less time consuming (Jargar et al 2012). This invention made us get The Physiological Society, UK scholarship to the University of Edinburgh.

Currently, we are working on isolation of specific bioactive compounds from plants origin of north Karnataka and elsewhere to develop drug(s) against ‘stroke’ and cardiovascular pathophysiology ( Kanthe et al 2021). Our Laboratory of Vascular Physiology and Medicine laboratory established the complex technique of rodent stroke model by middle cerebral artery occlusion (MCAO) technique. Our works on hypoxia preconditioning in cerebral ischemia as preventive measure made us internationally known by scientific and peer researchers (Bagali & Das,2021; Das et al 2021 ) Our laboratory is also working on computer-aided drug discovery through computational biology and artificial Intelligent (Parvatikar, et al 2021)

Me and my team also contributed a lot to environmental biology especially on groundwater arsenic and fluoride contamination in north Karnataka, India. He is one of the first to describe overt arsenicosis in such an area as well as in this region of Karnataka, India (Chakrabarti et al 2013). For this work, I had been conferred “Dr.Raja Ramanna State Scientist Award 2015” by the Government of Karnataka, India. https://www.kscst.org.in/awards_2013_2014/st_awards_2013_2014.html ). My contribution in hypoxia pathophysiology research is recognized with “Science, Honor & Truth Medal 2016” conferred by High Altitude Pulmonary and Pathology Institute, Zubieta University, Bolivia (https://www.altitudeclinic.com/award.html). Further, I had been conferred “G.P.Chatterjee Memorial Research Award 2019” by ISCA, Ministry of Science & Technology, Government of India (http://sciencecongress.nic.in/isca_awardees_2018- 19.php) for research on fluoride toxicities in Indi taluk of Vijayapur district, Karnataka, India (Ugran et al 2016). In 2021 for my work on COVID-19 in collaboration with the Republic of Bolivia, I was conferred the Plurinational Science and Technology Award 2021, awarded by the Ministry of Education, Government of Bolivia

INVENTION OF NEW METHOD TO DETERMINE BLOOD VITAMIN E (Α-TOCOPHEROL)

Invented   a novel procedure for specific and quantitative determination of α -tocopherol (vitamin E) in serum. Our modified method affords several advantages (i.e., simple, suitable, less tedious, economic, rapid, and utilizes a small quantity of serum) over existing methods (Das et al 2013).

The current modified method is based on the previous Baker and Frank method and the method of Martinek by using 2,2 ′ -bipyridyl, ferric chloride, and xylene. The complex of ferrous ions generated in this reaction with 2,2 ′ -bipyridyl is determined by using a plain enzyme-linked immunosorbent assay microplate (non-antibody coated) at 492 nm (Jargar et al 2012). This invention has brought to me “Shri Dev Raj Bajaj Memorial Research Prize 2014” conferred by the Association of Physiologists and Pharmacologists of India, Regd. office at AIIMS, New Delhi.

THE TECHNICAL SKILLS TO HANDLE BIOLOGICAL INSTRUMENTS:
  1. Expertise in handling different models of rats and mice as following:-
    • Chronic sustained hypoxia model (10% Oxygen; 90% N2)
    • Chronic intermittent hypoxia model (10% Oxygen; 90% N2)
    • Rodent stroke model by mid cerebral carotid artery occlusion(MCAO) technique
    • Protein restricted dietary model (5% Casein)
    • Hyperlipidemia dietary model in rats (30% fat)
    • Salt sensitive hypertensive (SSH) models by inducing L-NAME &NaCl
    • Renal arterial occlusion model for developing renal hypertension
  2. Well versed in handling Atomic Absorption Spectrophotometry to evaluate serum nickel and Cr VI concentrations as part of his research on metal toxicities in biological systems and environmental health research like groundwater arsenic concentration estimations.
  3. Experienced with working on electrophysiology setup in both humans and rodents. He is trained in handling ECG, EMG, EEG, and all the cardiovascular electrophysiological assessments including heart rate variability (HRV), small animal radiotelemetry devices, baroreceptor sensitivity tests (BRS) by using epinephrine and sodium nitroprusside techniques. These understandings are necessary for work on the hypoxia model in rats and mice.
  4. Experienced with the techniques of invasive blood pressure measurement through femoral/iliac artery of rats.
  5. Experience in central blood pressure measurements and arterial stiffness including augmented index and pulse wave velocity measurements through periscope and plethysmograph.
  6. Trained in ELISA, Western blotting, and PCR for quantitative and qualitative assessment for various Well-versed with various chromatographic techniques including HPLC.
THE PROFESSIONAL CAREER AS A PHYSIOLOGY TEACHER:

I am working in the area of biological sciences since his doctoral curriculum. I pursued Master’s and PhD in Human Physiology under University of Calcutta. I also did his Bachelor of Science degree with Honours in Physiology from Presidency College, Kolkata.

I have long standing academic and research career in biological sciences. I am teaching Physiology in medical school of India since 1989 for MBBS, BDS, MD, M.Sc students. I was also Visiting Professor of Physiology under Faculty of Medicine in School of Medicine, University of Leeds, UK (2014-2016) and Visiting Professor of Physiology in Tulane University School of Medicine, USA ( 2017).

I have successfully supervised/co-supervised 22 PhD and 10 MD (Physiology) students under Rajiv Gandhi University of Health Sciences, Karnataka, BLDE (Deemed to be University), Vijayapur, Vidyasagar University, West Bengal, KLE University, Belagavi and KIMS Deem University, Karad, India. I was the member of PG Board of Studies and PhD Registration committee of Rajiv Gandhi University of Health Sciences, Bangalore.

At present I am a Member of Education Committee of International Union of Physiological Sciences (IUPS). I am also the Vice President of The Physiological Society of India (2020-2023) and President of South Asian Association of Physiologists (SAAP) at University of Colombo, Sri Lanka..

THE VALUE OF MY WORKS IN THE PROGRESSION OF THE SCIENCES:

My research in basic biological sciences is extended to clinical medicine in general My works on heavy metals (nickel, chromium VI, lead, and cadmium) induced alteration of system biology are cited in WHO, EPA (ATSDR), etc. Our invention of a new and modified method of serum vitamin E estimation is widely cited and used in many diagnostic laboratories.  My team’s works on hypoxia biology also help to progress understanding of various respiratory diseases and uses of certain drugs like cilnidipine (calcium channel blocker) to treat chronic obstructive pulmonary disease. Further our works on plant extract to treat diabetes and the recent isolation of bioactive compounds from certain indigenous plant extracts to treat against cerebral ischemia or ischemic stroke is definitely fascinating and plays a pivotal role in the progression of medical sciences.

  1. I am currently holding the position of Dean of Environmental Health and Interregional Coordinator of UNESCO/UNITWIN Network under UNESCO Chair holder in Life Sciences (Biophysics, Biotechnology and Environmental Health) -2020 onwards till this day
  2. My team’s research was the first among the researchers in the world to report chronic human arsenic toxicity in any historic gold mining area (J Hazard Mater 2013 Nov 15;262:1048-55).
  3. Being myself a person with disability I work for disability issues with various NGO’s.
  4. At present I am working as the member of Education Committee of International Union of Physiological Sciences (IUPS), USA for the period from Jan 2015 to December 2018 and since 2018 to 2019 and 2022 to 2026.
  5. I was also holding the position of President of South Asian Association of Physiologists. University of Colombo, Sri Lanka (2020-2022) and currently in SAAP Executive body as immediate past president.
  6. I am a Vice President of the Physiological Society of India since 2014.
  7. I am also actively involved for the cause of Ethical Publication and am the Permanent Member of Committee on Publication Ethics (COPE), United Kingdom. Further I was also the first Indian to be awarded “COPE International Scholarship” at 2012 to present his works on ethical publications in London
